1. Examine Figure 5 on the FIGURES PAGE of the lab handout. Rearrange the steps in genetic
engineering in order from start to finish. Refer to the textbook page 425 figure 15-10 if needed.
2. Locate Figure 1 on the FIGURES PAGE of the lab handout. Referring to the steps you placed in
order, use a colored pencil to draw a thick line indicating where the restriction enzyme will cut the plasmid.
Remember: if you cut a circle, it can open up like a letter "C".

Cut out Figure 1 and tape/glue it in the appropriate space provided on the back of this sheet.
4. Locate Figure 3 on the FIGURES PAGE of the lab handout. Using a colored pencil, lightly shade in
the entire opened plasmid.

Cut out Figure 3 and tape/glue it in the appropriate space provided on the back of this sheet.
6. Locate Figure 2 on the FIGURES PAGE of the lab handout. Referring to the steps you placed in
order to know where to "cut", use a colored pencil to draw thick lines (more than 1) indicating where the
restriction enzyme will cut the human DNA.
7. Select a color you have not yet used to lightly shade in only the desired gene (the part between the
lines where the restriction enzyme will cut).
8. Cut out the desired gene from Figure 2 and tape/glue it in the appropriate space provided on the back
of this sheet.
9. Take a picture of your final plasmid (what you have on the desk in front of you after step 8 and place it at the
end of this document!!
