plasma membranes are very thin with a diameter of at most 10 nm. A good light microscope cannot resolve an image smaller than 2 micrometers so the plasma membrane was "invisible" to early researchers. Good electron microscope images were not available until the 1960s. Calculate how many times larger a 10 nm membrane would have to be in order to be viable under a microscope with a resolution of 2 micrometers.
